An Ambulance Victorian spokesperson said the man had been airlifted to The Alfred hospital with upper body injuries. He was in a serious but stable condition.
7.30pm
The man and woman injured in the crash were being flown to hospital in Melbourne, police confirmed.
Neither of the patients was said to be suffering from life-threatening injuries.
Police said they were starting to investigate the cause of the crash.
"All we can gather is they were heading east (on Duke Street) and something has happened," Central Goldfields Highway Patrol Senior Constable Norm Birthisel said.
"Whether it's a medical episode or something that has happened with the car, we don't know.
"There will be further investigation into the cause. We have to speak with other witnesses who saw the car go off (the embankment)."
