The incident occurred at an Axedale-Goornong Road intersection at Barnadown on Monday afternoon.
Police said the first driver, 34, was travelling along Epsom-Barnadown Road away from Bendigo in a white Ford Ranger when he failed to stop at a stop sign.
The car crashed into a white Volkswagen Amarok, travelling southbound along the Axedale-Goornong Road.
Police, Country Fire Authority crews and paramedics attended the crash.
An ambulance took the 34-year-old and the second driver, 66, to Bendigo Health for observation.
In May last year, the speed limit of the notorious Axedale-Goornong Road intersection was reduced from 100 kilometres an hour to 80 kilometres an hour.
The move followed a number of calls for action at the site, and came after a fatal crash in December 2017.
