Bendigo Health has said all testing would relocate to the Showgrounds on the day from 8.30am to 5.30pm.
"All testing will be carried out and children can get out of the car for a swab if they need to," a spokesperson said.
"There will be a priority lane for healthcare workers, patients, truck drivers and police."
They said McLaren Street will not be open for testing on Wednesday.
It comes as people were sent home from the McLaren Street COVID-19 testing hub on Tuesday after reaching capacity.
Queues started forming at the hub about 7am and numbers continued to swell throughout the morning.
Authorities allow for a two-hour wait period at the drive through and walk-in services for people lining up to be tested.
Bendigo Health is encouraging those who missed out on being tested to isolate at home if they have visited an exposure site.
