Bendigo Health chief executive officer Peter Faulkner said it was an important milestone to reach.
"There are 100,000 reasons why this is important for our community," he said in a social media post.
"Vaccination protects you, it protects your family, friends, our community at large and protects our healthcare system."
Mr Faulkner said every vaccination administered brings the population one step closer freedom.
"For those who have received their vaccination from brave teenagers to the most vulnerable elderly members of the community and everyone in between, thank you," he said.
"For all of our vaccination staff, you have done an amazing job creating a conformable and welcoming environment for our community and I thank you for your ongoing dedication."
Last week, Bendigo Health said they expected the region to reach the 80 per cent double-vaccinated milestone by the last week of October, well in time for an expected and much anticipated easing of restrictions.
Bendigo Health said it was based the projection on federal government weekly data published.
