Ambulance Victoria have confirmed a woman in her 30s suffering from chest and leg injuries was flown to the Royal Melbourne Hospital for further treatment.
They said a primary school aged girl suffering from facial and abdominal injuries was flown to the Royal Children's Hospital for further treatment.
A preschool aged girl suffering from chest and facial lacerations was taken by road ambulance to Bendigo Hospital.
All patients are in a stable condition.
The Loddon Valley Highway has re-opened in both directions between Worsley Road and Tragowel Road, Tragowel.
1.30pm:
Ambulance Victoria have confirmed two patients will be taken to Melbourne by two air ambulances following a road incident in Tragowel, near Kerang.
They said paramedics are currently treating a woman in her 30s for chest and leg injuries, a primary school aged girl for facial and abdominal injuries and a preschool aged girl for chest and facial lacerations.
A spokesperson said all of the patients are in a "stable condition".
Ambulance Victoria first received the call at 10.30am.
The Loddon Valley Highway is closed in both directions between Worsley Road and Tragowel Road, Tragowel due to a serious collision.
Motorists should follow the directions of traffic management on site and allow extra time for the detour using Leitchville-Pyramid Road and Boort-Pyramid Road.

Emergency services are currently responding to a road incident in Tragowel, near Kerang.
Ambulance Victoria have confirmed paramedics are currently treating three patients, one adult and two children.
They said the patients were being treated for "a range of injuries".
The spokesperson said an air ambulance had been sent to the scene.
Ambulance Victoria, Victoria Police, SES and the Country Fire Authority are all one scene.
