Woolcock, who turned 26 last Sunday on the day she defeated Indian Pranjala Yadlapalli for the first-week crown, opened her second week round robin campaign with a 6-3, 6-4 win over Elena Micic on Tuesday.
Her victory, which followed a bye on Monday, leaves Group B wide-open, with four players level on one win apiece after two days.
Jasmine Adams, who lost to Micic on day one, rebounded to defeat first-day winner Alison Bai 6-4, 4-6, 6-4 in a gripping three-setter.
New Zealander Katherine Westbury enhanced her chances of a berth in the semi-final round on Saturday by notching a second straight win to start the tournament.
The 27-year-old defeated Alexandra Osborne 6-3, 7-5 to be the only player in Group C with two wins.
Stefi Webb opened her account by defeating Zara Larke 7-5, 6-3, while first-day victor Monique Barry had the bye.
Barry and Westbury will clash in round three on Wednesday, presenting the 18-year-old with the chance to join her fellow New Zealander on two wins.
A finalist last week, Ydalapalli leads Group A with an unblemished 2-0 record following a 6-4, 4-6, 6-2 win over Amy Stevens.
It was the second-straight three-setter for Stevens, who overcame Rosin Gilheany on Monday 7-6, 1-6, 6-3.
Amber Marshall recovered from Monday's loss to Yadlapalli to beat Jessica Fowler 6-0, 6-0.
In Group D, 19-year-old Olivia Symons remained undefeated with a 6-3, 6-3 win against Kelsey Lewis, while Alicia Smith opened her account with a 6-4, 6-4 win over Mia Tsoukalas.
In the men's draw, Brydan Klein, who was a semi-finalist last week, solidified his bid for a return to the final four with a tough three-set win over Ken Cavrak.
Klein won 6-3, 3-6, 6-3 and moved to 2-0 following Monday's 6-2, 6-2 win over Jai Corbett. He is the only player in the Group with two wins.
He faces Alex Buite in Wednesday's third round, with Bulte coming off a second round bye and an opening day win over Thailand's Chancai Sookton-eng, who slipped to 0-2 with a two-set loss to Corbett on Tuesday.
Omar Jasika (C) and Adelaide's Sean Van Rensburg (D) head their respective Groups with unblemished records.
Jasika, the 2014 US Open boys' singles champion, chalked up win number two for the week with a hard-fought 6-3, 6-4 triumph over Corey Gaal, while Van Rensburg followed up his three-set win on Monday with a 6-1, 6-1 victory against Aidan Fitzgerald.
An intriguing Group B is up for grabs with four of five players on one win apiece.
Fresh from a first round bye, Dayne Kelly defeated Cooper White 7-6, 6-3, while Enzo Aguiard overcame Brandon Laubser 6-3, 6-3.
Following his bye on day two, Jeremy Beale (1-0) returns to action on Wednesday to take on White (0-2), who will be searching for his first win of the tournament.
