Searle Bros Building Contractors will perform the works, which the government described as a "major refurbishment."
They include demolition of a building, a hydraulic upgrade, a fire service and power upgrade, and a new building.
A library is among the new facilities, as well as multipurpose and meeting spaces.
College principal Steve Leed said the school had been preparing for the works for months, with classes and staff already out of the construction area.
There were makeshift arrangements for the library and multipurpose spaces, which are expected to be in place for about a year.
"The school community is really excited," Mr Leed said.
Works are expected to start soon after the school year ends, and to continue during the Christmas period.
The Christmas period is expected to be when significant work on the power upgrade takes place.
The state government this year announced $4.05 million for the project as part of its "school building boom".
The funding was additional to the $450,000 allocated in the 2019-20 state budget.
"Upgrading East Loddon P-12 College will give local students access to great classrooms and facilities," Member for Bendigo East, Jacinta Allan said.
