In the past week, Bendigo's three COVID-19 testing sites have conducted about 700 tests per day.
Ms Chesters said Bendigo needs a second Medicare-funded clinic to allow more people to access testing.
"The whole purpose of these clinics is to ensure people are being tested safely and quickly and to help prevent the spread of this virus," Ms Chesters said.
"The government should be doing more and it is disappointing they are dumping the burden back on the state's lap."

The Federal Government said a decision to open further respiratory clinics, like the GP-led Bendigo Respiratory Clinic in Spring Gully, is the responsibility of the state government.
A Department of Health spokesperson said the government is "supporting Victoria's response through a 50/50 cost-sharing arrangement under the National Partnership Agreement, which includes the sate led COVID-19 fever clinics."
The Bendigo Respiratory Clinic at 126 Spring Gully Road, Spring Gully is an appointment-only clinic, run by Spring Gully Primary Health, which assesses and tests symptomatic patients.
The government said the Commonwealth funded GP-led respiratory clinic servicing Bendigo has provided more than 5,200 assessments since opening in mid-April.

The clinic is conducting an average of about 100 tests per day and operates five days a week.
Demand for a COVID-19 test in Bendigo is expected to remain high, with Bendigo Health and the Department of Health and Human Services advising all families from St Joseph's Primary School in Quarry Hill be tested for COVID-19, according to principal Joan Coldwell.
On Monday, Bendigo Health's drive through testing clinic at Mercy Street closed early after reaching full capacity.
A Bendigo Health spokesperson said the drive through clinic has tested an average of 287 people per day in the past week.
The Stewart Street walk in clinic has seen an average of 295 people per day in the same period.
The GP-led respiratory clinic in Spring Gully was established to take the pressure off other GP clinics and hospital emergency departments, the government said.
There are 17 GP-led respiratory clinics in regional Victoria, including in Ballarat, Horsham and Warrnambool.
