One was taken to hospital after the incident with upper body injuries.
The Community and Public Sector Union has called for the Department of Justice to do more to protect workers at the facility.
The incident is the latest in a series of assaults against staff at the centre.
CPSU representative Julian Kennelly said an officer asked an inmate lingering at the fence line to move on.
Mr Kennelly said the inmate then spat at the officer and kicked him in the chest.
He said other officers were injured while restraining the inmate.
Mr Kennelly said the union was in talks with the Department of Justice.
He called for some offenders to be moved away from the Malmsbury site.
"We're extremely concerned about the health and safety of the staff working at the Youth Justice Centre and Department needs to do more," Mr Kennelly said.
A Department of Justice spokesperson said violent behaviour or assaults at youth justice centres were absolutely unacceptable.
The spokesperson said the safety of staff and young people was of utmost importance.
"New laws are now in effect that strengthen consequences for young people who assault youth justice custodial workers, damage property, and escape or try to escape," the spokesperson said.
"This includes a presumption they will serve their sentences on top of an existing period of detention."
