The event is held in up to 100 locations across Australia, with Bendigo hosting its local event for the 11th time.
Participants can opt for a four kilometre walk or run, or an eight kilometre run.
Taking place at the relatively flat Beischer Park in Strathdale, distance runners will complete two circuits of the picturesque track, while taking in the beauty of the local bushland park and Kennington Reservoir.
The four kilometre participants will only need to complete one circuit.
Established in 1998, the Mother's Day Classic aids breast cancer research.
Every dollar raised by the event goes to the National Breast Cancer Foundation to fund its breast cancer research.
In 22 years, there has been more than $37 million raised for cancer research, spread across 70 research projects.
Jody McGregor is organising the Bendigo Mothers Day Classic for the second time and said breast cancer has touched everyone.
"It is something that is close to my heart.
"To honour and remember people touched by breast cancer makes this a unique event," Ms McGregor said.
The 2020 Mothers Day Classic will feature face painting and a coffee van to improve the experience for spectators.
Prior to the event, a powerful minute's silence is held to commemorate those who have been touched by breast cancer.
"Last year was my first time organising the Bendigo event and that minute's silence was amazing.
"Remembering and celebrating those that have been touched by breast cancer was quite humbling," Ms McGregor said.
Participants are encouraged to set up their own team fundraising pages, dress up and submit corporate and school teams, with a prize on offer for the largest participating team.
The Mothers Day Classic is looking for volunteers, particularly to help facilitate the day's events.
