A passing motorist found the man, 55, on Fogartys Gap Road at Ravenswood South about 11.30pm on Sunday.
An Ambulance Victoria spokesperson said the Maldon cyclist was flown to the Royal Melbourne Hospital in a critical condition, with upper body injuries.
"Investigators are yet to establish how the man sustained his injuries," police said.
"[We] wish to speak to anyone who may have been travelling along Fogartys Gap Road and saw [the] cyclist prior to police arrival."
Police urged anyone with information to contact Crime Stoppers online, or call 1800 333 000.
The rider was found on the same section of Fogartys Gap Road where a motorist was killed in a single-car crash just before Christmas last year.
It is 14 months since the cyclist Michael Grinter was struck and killed by a car in a separate crash on the road.
The driver was jailed for three years after pleading guilty to causing the crash.
