Highlights include the Eaglehawk Heritage Precinct, encompassing the Mechanics Institute, the former Eaglehawk Town Hall and the Eaglehawk Courthouse, with its original log lock-up.
Architecture on show during the weekend extends throughout Bendigo, even taking in Harcourt and Castlemaine for the first time.
Bendigo's historic mining exchange will be open to the public. Organisers say the public will have a new level of access to the exchange - known as the Beehive building - after it was part of the program last year.
The building is due for completion in just six weeks, after four years of development.
Sustainable, and budget friendly, home Koornook House II will also open, giving viewers an insight into its design.
Open House Bendigo program and business manager Victoria Bennett said event chose sites that were significant, or remodelled to a new use. She said Eaglehawk was an unusual precinct, showing well maintained and original heritage features.
Open House looked for buildings pushing design boundaries when it came to contemporary architecture, Ms Bennett said.

Ms Bennett said visitors would get access to the Beehive just before it opened to the public in its new form.
The public only had access to the ground floor at the 2018 Bendigo Open House.
Ms Bennett said the upper floor access would give people a full sense of the building.
