The Team Sunweb rider is 21 seconds behind leader Patrick Bevin, with reigning champion Daryl Impey in second place just seven seconds off the lead.
Aussie star Richie Porte is alongside Hamilton 21 second behind.
Hamilton, who leads the young rider section, and Porte will both be suited by Sunday’s gruelling summit finish at Willunga.
Porte has won that stage the past five years.
Impey took out Friday’s fourth stage with a well-timed run.
Impey dedicated his win to family friend Justin Bitter, his wife's cousin, who died earlier this month from a heart attack.
Impey (Mitchelton Scott) aims to become the first rider in the Tour's 21-year history to successfully defend his title.
Bendigo rider Jason Lea still leads the King of the Mountain classification.
Lea has 20 points – four clear of Kiwi George Bennett.
