Ambulance Victorian confirmed a man in his 50s was taken to the Royal Melbourne hospital via road ambulance with injuries to his face and head.
He was in a serious condition.
The other person taken to hospital was a man in his 60s, but his injuries or condition were unknown, according to an Ambulance Victoria spokesperson.
Northbound lanes of the Calder Highway are closed at the Edgecombe Street overpass.
Traffic is currently being diverted off the highway at the Edgecombe Street exit ramp then back on onto the highway at the on ramp.
