The 38-year-old male suffered leg injuries and was taken to Bendigo Health in a stable condition, an Ambulance Victoria spokesperson said.
The four-wheel-drive the man was driving rolled on a straight stretch of the Calder Alternative Highway about 4.30am.
Acting Sergeant Paul Stubbs said police were unsure at this stage as to what led to the crash but further inquiries were being made.
