It is believed two utility vehicles travelling in opposite directions along the Calder Highway, approximately 2km north of Adams Road, collided head-on shortly after 2pm.
One of the vehicles came off the road and caught fire following the collision.
The driver, who is believed to be the single occupant of the vehicle at this stage, died at the scene.
The deceased is yet to be formally identified.
A male and female, who were the two occupants of the other vehicle, were treated on scene and conveyed to hospital with non-life threatening injuries.
The Calder Highway between Yeungroon-Woosang and Yeungroon roads is currently blocked to traffic in both directions and diversions are in place.
The exact circumstances surrounding the crash are yet to be determined and the investigation is ongoing.

UPDATE 3.45pm: The Calder Highway is closed to traffic south of Charlton at the Yeungroon-Woosang Road and north of Charlton at Yeungroon-Charlton Road.
EARLIER: Emergency services are on scene at a multi-vehicle crash near Charlton.
There are reports one car caught fire following the crash on the Calder Highway.
Ambulance Victoria was called to respond to three patients shortly after 2pm.
