Holmes, one of the stars of the Bulldogs' 2015 premiership win over Strathfieldsaye, was unveiled as coach on Tuesday.
She replaces Lia Marrone, who coached the Bulldogs in more than 100 games, but did not seek reappointment.
Marrone, who was a player in the Bulldogs' A-reserve premiership win last season, was coach of the club's 2015 premiership team.
She also led them to a grand final berth in 2015.
Holmes takes charge of a talent-laden Golden Square line-up, which finished runners-up to Kangaroo Flat in last season's premiership race.
The defender split time during the 2016 season with Golden Square and Cohuna Kangas in the Central Murray FNL.
She was best-on-court in the Bulldogs’ 2015 flag win, a year in which Holmes polled 21 votes to finish fifth in the league best and fairest Betty Thompson Medal count.
Golden Square vice-president and secretary Brendan Stewart, who was part of the selection committee alongside Michelle Ryan and Phil Delahunty, welcome the appointment of Holmes.
"Allira is well known to us as a champion player and she has recently secured teaching work in Bendigo for the 2017 school year," he said.
In other coaching appointments, Melinda Keighran will coach the Bulldogs' 17-and-under team, after spending last season with the AFL Central Victoria Academy.
"She brings a whole new skill set to our developing netballers," Stewart said.
Further coaching appointments, including the Bulldogs' A-reserve, B-grade and C-grade teams are expected after a club meeting on Thursday night.
Despite securing only one premiership, it was a successful last season for the Bulldogs.
The club's A-reserve team made it a premiership three-peat with a big win over Sandhurst, while the Bulldogs' A-grade and B-reserve teams played off in grand finals.
Meanwhile, try-outs for the Bulldogs' 17-and-under squad get underway this Wednesday, with registrations from 5pm ahead of a 5.30pm start.
Further trials will be held on Tuesday, November 8 and Thursday, November 10, 5.30pm start.
Senior tryouts will be held on Tuesday, November 22 (registration at 6pm), Thursday, November 24, and Wednesday, November 30, starting 6.30pm.
