Heathcote Bowling Club is hosting the unique Australia day celebration, which rolls into action at 3pm Monday.
The aim is to have at least one game of bowls being played on the club's Herriot Street greens non-stop for 24 hours.
Club stalwart Brian Taylor said MND was a cause close to the hearts of all club members after one of their teammates was diagnosed with the condition last year.
"We considered a 24-hour marathon was one way we could involve the district community in fund-raising, as well as helping people to understand more about the disease," he said.
Bowlers will donate $1 for each end of bowls they complete during the 24 hours.
The event has attracted support from players in Colbinabbin, Geelong, Gisborne, Rochester and the Yarraville-Seddon Bowling Club.
Teams led by federal Member for Bendigo Lisa Chesters and state Member for Euroa Steph Ryan will also take to the greens, alongside a group of teachers from the Kennington Primary School.
The staff members will play in honour of former teacher Mark Winter, who lost his battle with MND last October.
MND Victoria has thrown its support behind the event, as have local businesses, which have donated items for raffles and a trading table.
Openings remain in some time slots, with Mr Taylor encouraging players and non-bowlers to take part.
Motor neurone disease (MND) is the name given to a group of diseases in which the nerve cells, or neurones, controlling the muscles that enable us to move, speak, breathe and swallow undergo degeneration and die.
The condition causes weakness in the muscles that gets worse and eventually leads to paralysis. There is no cure.
