Will Green and James Dann will run from Melbourne to Bendigo in support of the Sidney Myer Foundation - a Haven; Home, Safe initiative.
Haven; Home, Safe is a Bendigo organisation that provides services to the homeless and disadvantaged.
The Sidney Myer Project aims to break the cycle of homelessness in Bendigo.
The project will see the development of a 23-unit medium-term residential complex for singles, couples and families and provide them with a development program to help them become engaged in the community.
Charter Hall, which owns Lansell Plaza, is undertaking a $38 million renovation of the shopping centre.
Through the run, the company will set out to raise $25,00 for the project.
Mr Green said the 132-kilometre run would prove a difficult challenge but the end result would make it worthwhile.
"We wanted to do something significant that will not only allow us to raise funds but also raise awareness of this important project," Mr Green said.
"The Sidney Myer Haven project will make a significant difference to the lives of a number of families in crisis. James and I are proud to be a part of this event and give back to a community who supports us."
The Sidney Myer Haven residential complex will be built on the former nursery site in Somerville Street, two kilometres from Bendigo's central business district.
Haven; Home, Safe chief executive Ken Marchingo said the development would be a pioneer for the future delivery of crisis accommodation and homelessness programs in Bendigo.
"Program participants will be seeking to make a change in their lives and be prepared to move out of their comfort zone," Mr Marchingo said.
Mr Green and Mr Dann will run from Melbourne to Bendigo between November 18 and November 20.
The duo will write updates on their training in the lead up to the run on the Lansell Square Facebook page.
