Dr Napthine said the government had a clear policy of trying to create jobs in regional Victoria.
"In the last 12 months we've created over 25,000 new jobs in regional Victoria," he said.
"We're keen to grow jobs in regional Victoria.
"These things are speculated on ... it's premature to make a comment on it at this stage."
If the department's headquarters were relocated to Bendigo it would signify a big jobs boost to the region.
In March the state government announced that the VicRoads headquarters would be moved to Ballarat if re-elected to power.
He said this would pump $40 million to the local economy and would be a "win-win" situation.
