Both line-ups are chasing their first points after first-round defeats.
Bendigo's line-up is on the road again for the match on the Rangers turf at Epping.
The day's play kicks off with the under-20s at 4pm, and the seniors from 6pm.
Coach of FC Bendigo's senior team, Steve Martin said some changes had been made to the line-up which was beaten 3-2 by Avondale Heights at Thornbury.
Viktor Rodriguez will take his place in Bendigo's midfield.
"Viktor is an attacking central midfielder, is really creative, and has some experience from playing in Spain," Martin said.
"Josh Romein from Ballarat Red Devils is in the defence."
After two weeks training since its debut match, FC Bendigo has worked hard on skills.
"There were a couple of lapses in concentration which cost us in round one," Martin said.
"It's important we learn from those mistakes. It's not just about missed passes, but also about structures and putting pressure on the opposition."
Josh Naunton and Jono Ryan have been ruled out after they collided at Thursday night's training session.
FC Bendigo captain Trent Waterson will again play a key role in midfield or attack.
"Trent's pace and attacking mindset add some X-factor to our team," Martin said.
FC Bendigo's inaugural home match is on Saturday, April 5 against Moreland Zebras at Epsom-Huntly from 7pm.
