Sergeant Tony Commadeur said police were still investigating the incident, but said the men may have been glassed.
“It occurred just on midnight,” he said.
“One male received injuries to his face and body.
“Another male also received injuries to his face.
“Both have ended up in hospital.
“One of them has received serious injuries and will be in hospital for some time.”
Sergeant Commadeur said early indications were the men had been innocent bystanders who had tried to stop the fight.
“A large number of people were involved in the incident,” he said.
“Whether it was an all-in brawl or a whole lot of people standing around, I don’t know.
“It’s in the hands of investigators.”
The victims were from Heathcote and Melbourne, he said.
The age of the victims is not known.
A spokesman for the One Tree Hill Hotel said a group of patrons left the pub about midnight.
"As our staff were cleaning up they were alerted to an argument outside the pub near the roundabout," he said.
"There was a large group arguing and the police were instantly called.
"A fight broke out near the same spot and by the time police arrived, the fight was broken up by our staff and the police took over from there.
"The group involved were allegedly not from the area and were well behaved inside the venue."
Police will request CCTV footage of the incident from the hotel, which has several video cameras.
It's believed police have not questioned any suspects over the matter.
Anyone who witnessed the incident should call Bendigo Police on 5448 1300 or Crime Stoppers on 1800 333 000.
