Police are currently on scene following the incident, which occurred just after 3.30pm today.
"A Mazda sedan was travelling south on Mount Macedon Road when it collided with a motorcycle," a spokeswoman said.
"The male motorcyclist, who isyet to be identified, died at the scene.
"No one else was injured.
"The driver of the Mazda, a Woodend woman, is helping police with their inquiries."

3.50PM: A PERSON has been knocked off their motorbike in a collision with a car on Mount Macedon Road.
Ambulance crews are now at the scene.
Ambulance Victoria spokesman John Mullen confirmed they received the call at 3.26pm.
“It is between a car and a motorcycle,” he said.
“The helicopter has been paged.
“The crew have only just arrived on scene and they will assess the situation.”
