Bendigo East MP Jacinta Allan formally announced works began on Friday.
Upgrades will widen the intersection, install new traffic lights and improve street lighting.
Works also include a new shared footpath and dedicated bike lane to keep pedestrians and cyclists separated from drivers.
Strathfieldsaye Road is used by more than 9000 vehicles a day to access local schools, businesses, health services, pre-schools and sports facilities.
During upgrade works, which are expected to be complete by mid-2018, some closures may be required over summer to complete the project.
Transport Minister Jacinta Allan said: “We’re making Strathfieldsaye road safer and less congested for local students and families.”
“While there will be disruption in the short term, this major upgrade will boost traffic flow and safety for decades to come.”
VicRoads began preliminary works on the intersection of Strathfieldsaye Road, Tannery Lane and Club Court, in October.
