The Magpies produced one of their best wins in recent memory to defeat the Roos 52-39 in a tough contest at Princes Park.
Goal shooter Jane Reid was in strong form for the Magpies, scoring 30 goals.
But it was the efforts of defensive trio Eliza Roughead, Chloe Egan and Ash Lierich that won most praise from coach Anika Munn.
"It was a very tough game, but our defenders were very good," she said.
"They played a ripping first half and really shook the confidence of the Kangaroo Flat goalies and we were able to convert from their turnovers."
The Roos were without star goal shooter Ruby Barkmeyer, who will next week represent an Australian junior team in Fiji.
Meanwhile, Golden Square roared back to form with a vengeance after last round's loss to Sandhurst to defeat fellow finals contender Eaglehawk at Canterbury Park.
The Bulldogs posted their third win from four games with a stirring 57-40 victory on the Hawks' home court on Saturday.
Sandhurst has maintained its undefeated status following a 78-22 win over Kyneton.
South Bendigo continued its good form with a 29 goal win over Castlemaine. It was the first time the Bloods have strung together back-to-back wins since rounds 17 and 18 of the 2004 season.
Gisborne rebounded from a tough loss at Kangaroo Flat to defeat Strathfieldsaye 52-29.
