The hardware chain is putting the final touches on the $27 million store and is expected to open to the public in the middle of June.
Many of the new team members are qualified tradespeople, including carpenters, plumbers and electricians.
Bunnings Warehouse Epsom complex manager Paul Noel said a majority of the new staff came from the City of Greater Bendigo area and would be joined by about 30 current staff.
He said the opening of the new store was exciting.
"Bunnings provides a wide range of meaningful career development options for all team members and I am confident our new recruits will relish the opportunity to learn and grow with us," he said.
"Bendigo has needed a new Bunnings store for a long time. It's exciting."
Mr Noel said the store meant people in Epsom and surrounding areas would not have to travel to Kangaroo Flat to go to Bunnings.
"Potentially there are a lot of new customers who will venture to our store," he said.
Bunnings Warehouse Epsom activities organiser Kirsty Patterson said new team members would spend time in the community over the coming weeks.
"Giving back to the community is a huge part of what they do - we're very community-minded," she said.
Ms Patterson said staff members regularly dropped into schools, kinders and other non-for-profits to conduct community activities, from gardening to DIY projects.
"We now have two stores in Bendigo which is pretty big for a regional town. This means we will have even more staff working in the community. It's really great and exciting," she said.
