The star from Bendigo Harriers raced to victory in a time of 8:33.48 at the Latrobe University Bendigo athletics complex in Flora Hill.
The previous best of 8:33.7 was achieved by Leigh Purtill on December 7, 1985.
It was a superb run by Threlfall in preparation for Saturday night's running of the Flack Advisory-backed Richard Kitt Memorial 1500m at the Bendigo International Madison carnival.
Threlfall will be co backmarker with Andy Buchanan off 1.55 in the AB classic to be run at the Tom Flood Sports Centre in Barnard Street.
