Both clubs have featured in the past two grand finals in which Bendigo won both times on its court.
Spirit's quest for a premiership hat-trick has struck a few obstacles this summer.
Coach since the club's formation eight years ago, Bernie Harrower said playing in Townsville would be a "huge challenge" for the Australia Pork-backed Bendigo Spirit line-up.
This season's record between the teams stands at 2-1 in Bendigo's favour, but Fire tipped off the season by winning the round one clash against Spirit at home by three points.
The next two contests were in Spirit's territory where Bendigo won by three and then 11 points.
"We have the game-plan and the players to get the job done," Harrower said.
"Defensive play will be crucial.
"Townsville is a high-scoring team, so the key is clamping down on their attacking moves," Harrower said.
At the other end of the court, Bendigo has the scoring punch to trouble Fire.
Kelsey Griffin was MVP in the past two grand final wins and teams brilliantly with the likes of Kelly Wilson, Gabe Richards, Sara Blicavs, Belinda Snell, Maddie Garrick and Tessa Lavey.
"Our senior leaders have handled adverse situations really well the past six weeks," said Harrower.
Harrower said hosting a final added to the expectations.
"Townsville's been the dominant team all season, so the pressure on them is greater.
"As a group we are not daunted by playing in Townsville. We are excited by the challenge and the chance to win our way to a third grand final in a row."
After being on the sidelines last round because of injury, Spirit guard Kelly Wilson is right to play.
WNBL play-offs action starts at 3pm on Saturday when Dandenong plays Sydney.
The Townsville Fire and Bendigo Spirit semi-final match tips off at 3pm (AEDT) on Sunday and will be televised on ABC2.
