Seven people were killed and 76 seriously injured on Victorian roads over the three day period last year.
Bendigo Highway Patrol Senior Sergeant Mark Edwards said police would be out in force, saturating roads.
Operation Amity, running from January 23 to 26, will target speed, drink and drug driving, distraction, fatigue and seat belt offences.
The operation will place particular emphasis on motorcyclists after riders sustained about a third of all serious injuries recorded last Australia Day weekend.
Two motorcyclists have been killed in Victoria this year, including a man at Kyabram on January 1.
It is believed the man, 35, was not wearing a helmet.
Senior Sergeant Edwards said bikers could stick to the speed limit and still enjoy their ride.
VicRoads have echoed the Victoria Police warning, urging motorists to avoid drink driving.
"Whether you are just driving around the corner for a BBQ with friends and family or travelling further afield to celebrate this long weekend, please plan to arrive alive," VicRoads regional director Mal Kersting said.
"If you plan on drinking, don’t plan on driving. The best thing you can do is arrange alternate transport.
"If it's a short trip, why not take a walk or a taxi ride.
"For those longer journeys try public transport or nominate a designated driver to ensure you get to and from your destination safely and can enjoy your long weekend."
