UPDATE 10.10pm: The 80-year-old woman who was a passenger in the car remains in a critical condition in The Alfred Hospital in Melbourne.
The driver of the truck was released from Bendigo Health in the afternoon following observation.
UPDATE 4.55pm: Police have said the Calder Alternate Highway is still closed as the Major Collision Investigation Unit and Heavy Vehicle Unit continue their investigations.
It is unknown when the road will re-open.
UPDATE 2pm: Police have confirmed the pair travelling in the car were from Maryborough.
The passenger, who died at the scene, was a 40-year-old woman, police said.
The driver, who was flown to Melbourne with life threatening injuries, is an 80-year-old woman.
Police said it appeared the car failed to give way to a truck travelling south on the Calder Alternate Highway.
UPDATE 12pm: The "elderly" woman flown to Melbourne was the driver of the car, police have said in a statement released to media.
She has life-threatening injuries and was travelling in the car with one passenger, who died at the scene.
Police are continuing to investigate the circumstances surrounding the crash, which officers say happened about 9.35am.
Police are urging anyone who witnessed the crash or may have seen the car before the collision to contact Crime Stoppers on 1800 333 000 or submit a confidential crime report at www.crimestoppersvic.com.au
The provisional road toll for 2015 is 17 compared to 12 in 2014.
UPDATE 11.25am: A female passenger who was travelling in the car has been flown to The Alfred hospital in Melbourne.
She was travelling in the car with one other person, who police earlier confirmed had died.
The truck driver has been taken to Bendigo Health.
Police believe the car was travelling along the Bendigo-Maryborough Road and collided with the truck as it attempted to cross the Calder Alternate Highway.
The truck was travelling south-bound on the Calder Alternate Highway, police said.
The Major Collision Investigation Unit and Heavy Vehicle Unit are both on their way from Melbourne.
Traffic blocks will remain in place for at least another four hours, police said.
UPDATE 10.55am: Paramedics are loading someone into the air ambulance.
Air ambulance has left the scene.
UPDATE 10.50am: A Lockwood resident said the Lockwood Road crossing is a notoriously dangerous road.
The man, who did not wish to be identified, said he has lived in Lockwood for 11 years.
"I travel this road every day and you need eyes in the back of your head," he said.
"The big trucks come flying through here pretty fast and you must always stop and look.
"Things like this happen if you are not careful. You must be careful."
UPDATE 10.30am: One person has died, a police officer on scene has confirmed.
The collision involved a truck and a car at the intersection of Lockwood Road and the Calder Alternate Highway.
Air ambulances have arrived on scene.
North bound traffic on the Calder Alternate Highway is being diverted up Crusoe Road.
South bound traffic is being turned back to Marong.
Lockwood Road traffic is being diverted back to Bendigo.
The car involved in the collision, a Holden Camira, is lodged under the truck.
The SES has removed a door of the car.
UPDATE 10.05am: The crash involved a car and a B-double truck.
SES crews are currently working to free a person from the car.
There are currently four paramedic crews, two CFA tankers police and SES crews on scene.
Traffic is being re-routed around the intersection.
South bound traffic on Calder Alternate is getting redirected up Lockwood Road.
UPDATE 10am: An Ambulance Victoria spokeswoman said the crash involved a car and a truck on the corner of the Calder Alternative and Lockwood Road.
She said ambulance were assessing two people on scene.
