The south-bound approach from the Calder Alternative Highway will also be reduced to 80 km/h.
The changes are part of a set of safety measures while VicRoads prepares to build the new interchange.
Other measures on the Calder Alternative will include new rumble strips, new electronic message signs, larger warning signs on all approaches to the intersection, greater visibility of Stop signs and more prominent line marking.
Construction of the new interchange will begin in late 2015 and include two bridges to carry traffic over both north and south-bound lanes of the Calder Highway.
The speed reductions will remain in place until after the new interchange is completed in 2017.
The $86 million project, made up of state and federal funding, received bipartisan support in August last year.
The speed on an 800-metre stretch was reduced to 80 km/h in February.
