President Lisa Maud said the Golden Square facility had been marked with spray paint during the early hours of Friday morning, with one of the markings reading 'Jack is back'.
She said the attack comes after City of Greater Bendigo funded a refurbishment of the building - which included a fresh coat of paint on its exterior - just over six months ago.
"It's extremely disappointing," she said.
"There is clearly someone out there with too much time on their hands."
Ms Maud said the club had installed security fencing worth $16,000 about three weeks ago to prevent vandalism and believed the offenders had gained entry via a public access entrance - a requirement on council land.
She said the club did not have CCTV surveillance cameras.
"I think that is something we will be looking at getting in the future," she said.
"We will be trying to get a grant to fund some security cameras so that this doesn't happen again."
Ms Maud said she was unsure how difficult it would be to remove the graffiti but hoped it would be complete by tomorrow, with the club scheduled to host a car boot stall.
"It stands out a lot because everything is so new," she said.
"It's really disappointing that this has happened."
