Bendigo's Sergeant Mark Dole said police pulled the vehicle over about 5.30am.
He said police found what was believed to be marijuana before a thorough search of the car revealed quantities of what was believed to be ice and cocaine.
He said the driver was found to be disqualified.
He said the men, all from Bendigo, were taken to the station and interviewed.
He said they had all been released and would be charged on summons once an investigation of the matter was completed.
Sergeant Dole said it was rare to find cocaine in central Victoria because of the expensive price.
"This is the first time I can remember cocaine being located (in Bendigo)," he said.
"Others members say they have rarely seen it in the past."
