The 35-year-old was travelling along the Castlemaine-Maldon Road towards Castlemaine when her Mazda 3 sedan veered off the road and hit a tree.
The incident happened about 1.15pm.
She was the sole occupant of the vehicle.
Castlemaine police Sergeant Rohan Scott said the cause of the accident was unknown but police had ruled out speed, alcohol and drugs.
"There is no apparent cause of the incident," he said.
Sergeant Scott said paramedics treated the woman on scene.
He said it was a reminder for drivers to take care on the roads.
"Given the Easter break we urge country drivers to concentrate on the roads," he said.
