Among those chipping in to back Brennan's trek are Bendigo Spirit stars Gabe Richards and Kelsey Griffin.
Brennan was joined by the dual WNBL championship winners for Wednesday's promotion of the run at Bendigo Marketplace.
Friday's run is all about raising funds for Ovarian Cancer Australia.
A key off-court player for the Spirit as fitness advisor, Brennan starts his trek from Echuca at 6.30am on Friday.
He is aiming to complete the 92km trek by 6.15pm when he arrives at the Chinese Gardens in Bridge Street.
There will be little time for Brennan to rest as South Bendigo plays Balwyn Tigers in Saturday's match for the Shaun Bergin Cup.
The Bloods and Tigers will play at Epsom-Huntly Recreation Reserve.
The match honours the skilful wingman who died after being injured in an on-field clash playing for the Tigers in September, 2011.
Bergin also played for the Bendigo Bank Pioneers in the AFL Victoria TAC Cup under-18s competition and was a rookie-list player with Hawthorn.
Saturday's footy action at Epsom-Huntly starts with the reserves at 10am.
The main game starts at noon.
