It was back-to-back wins for Coles who a week earlier won the Sens Jewellers Wheelrace.
In Thursday night's showdown, Coles began from a mark of 110 metres in the 2000m event on the Barnard Street track.
From the backmark of 90m, Jade Maddern looked the likely winner at 300m to go, but Coles and Toby McCaig (160m) swooped around the final bend and along the straight to be first and second.
Next across the line were Scott Keating (140m), Alessia McCaig (130) and Belinda Bailey (180).
The first of the A-grade scratch races was a 10-lap duel in which Coles, Jade Maddern and Emerson Julian were the placegetters.
In the A-grade 15-lap motorpace it was Maddern who was too strong for Coles, Scott Keating and Toby McCaig.
Senior racing began with the DCK Real Estate-sponsored Crystal Classic women's wheelrace.
The Humber sisters, Iesha and Alaya achieved a one-two result from marks of 50m and 85m.
Next best were Leshae Maddern (110m) and Milana Freer (115m).
In the juniors action, Tobias Jelbart marked three wins from as many starts in the under 11-13 events.
Other rising stars in form included Lilyth Jones, Bryce Nicholls, Lewis McIntosh, Heidi Larson and Poppy Maw.
Meanwhile, the 85th Melbourne Cup on Wheels was raced on Saturday at the Darebin velodrome.
Bendigo's Ned Pollard raced from the 120m mark and was third in the classic as Ballarat's Daniel Bucknall (125m) and Blackburn's Sean Cleary (195) led the charge.
A multiple national track champion, Bendigo's Alessia McCaig began on the 10m mark in the women's Melbourne Cup on Wheels final.
McCaig was runner-up to Brunswick's Amelia Mulhern (150m) as clubmate Fiona Bonnett (170m) was third.
Track racing rolls on in Bendigo this Thursday night.
Juniors start at 6.30pm and seniors from about 7.15pm.
Main event for the senior riders is the Profound Civil Contractors Wheelrace (2000m).
Another heat in the Bendigo Athletics Club's distance series will be run.
