The Strathfieldsaye testing site has now closed off the line for the day.
Staff have requested police assistance and traffic management to assist with the volume of cars waiting.

The queue for testing extended from Aspley Street, up Uxbridge Street and along Strathfieldsaye Road before extending down Club Court.
"We didn't expect numbers to this extent," he said. "We have had a lot before but not like this where it has gone all the way around the corner.
"The Bendigo Health testing sites have already started turning people away and we are nearly at that point."
"It becomes a very stressful day, especially knowing it affects traffic as well. Patients have been waiting for a few hours in the car. It's not easy on them if they need toilet breaks, lunch breaks or things like that. But we have to pull together."
The increase in testing across Bendigo comes as a number of retail stores in the CBD and Bendigo Marketplace were listed as exposure sites.
On Monday morning, COVID response commander Jeroen Weimar confirmed a new case in Mount Alexander was linked to the Bendigo exposure sites.
Dr Ordukaya said testing numbers as the site fluctuate depending on when there are exposure sites.
"Days like this is goes into the two hundreds but it fluctuates depending on when there is exposures," he said.
"We do as many as we can with the pathology COVID testers. We have managed to get a few more on today but if we had more testers we could see more.
"We're a GP clinic and the Clinical Labs are doing the testing. We have myself allocated for COVID testing and other doctors seeing patients."
"We are urging the whole Bendigo community to be patient and be nice to each other. We do get people having a go at the staff. Just remember everyone is under the pump, so be patient and we thank them for doing the right thing."
