Victorian chief health officer Brett Sutton confirmed both cases recorded overnight have what's known as the Delta variant of COVID-19.
"The two cases from yesterday are quarantining in hotel quarantine and we are providing support to them," he said.
Health Minister Martin Foley said the latest cases were exactly why it was vital for Victorians to remain vigilant, wear masks indoors and check in everywhere when visiting venues.
"It is why everyone should get tested as soon as they have the slightest of symptoms," he said.
"Our public health teams have taken the strongest of actions to wrap around these cases and I want to thank them for that.
"It is up to all of us to play our part to keep each other, our families and our community safe especially when we come to terms with this delta variant of concern.
"It moves faster, it's more infectious and it knows no boundaries."
It comes as NSW Premier Gladys Berejiklian announced four Sydney local government areas would go into lockdown after the state recorded 22 new locally acquired cases of COVID-19.
There are now 65 cases linked to the Bondi cluster.

VICTORIA has recorded two new COVID-19 cases in the 24 hours to midnight.
The two cases were already announced Thursday afternoon and include a man who returned from Sydney on Sunday night and his co-worker.
The first case is a man in his 60s who attended a social event at his daughter's house in West Hoxton last Saturday, which was at the time a Sydney green zone, and then returned to Melbourne on Sunday.
He developed symptoms late on Tuesday night at his home in Oakleigh, where he lives alone, and was tested on Wednesday.
Primary close contacts of the case are being identified and contacted and a workplace exposure site linked to this case, a dry-cleaning business in Sandringham.
This case's only workplace close contact has received notification of a positive test.
His family has been isolated and are being tested, and urgent public health actions are underway to understand the case's movements.
These cases take the number of Victorian cases linked with the growing Sydney outbreak to two.
A total of 18,019 people were vaccinated and another 21,991 were tested for the virus.
New exposure sites have been added in Victoria for the first time in more than a week and include Melbourne Airport, Flinders Street and Broadmeadows train station.

Restrictions across the state have now eased meaning Victorians are now allowed to have 15 visitors at their place per day and hold public gatherings of up to 50 people.
Masks remain mandatory in indoor settings outside the home, and are recommended outdoors when social distancing can't be maintained.
Hospitality venues can now serve up to 300 in Melbourne, but density limits still apply.
Restrictions on hospital and aged care visitation eased.

It is a far cry from the situation in New South Wales, where 11 more active COVID-19 cases were added on Thursday morning.
The state's premier Gladys Berejiklian said it was the "scariest period" NSW had faced since the start of the pandemic.
