Set to reopen its doors in August, the winery at Big Hill will join the company's already established vineyards and cellar doors at Echuca-Moama, Myrniong and Lorne.
St Anne's Winery owner and chief winemaker Richard McLean said the team had been keeping their eyes on the region for a good while.
"We've been looking for new winery sites for a number of years and we've looked at a few around Bendigo as well as other places," he said.
"This place popped up at the start of the year and it seemed like a pretty good fit for us so we jumped at the chance."
But this isn't the first connection the company has had with the area.
"We already grow and make a number of wines from the Bendigo region so we quite liked the wines from the area, the wines we make are quite successful," Mr McLean said.
He said they had big plans for the 95 acre site.
"We will definitely be having a cellar door at the Bendigo site and will be selling our full range of wines,' Mr McLean said.
"We'll be doing lunches there on weekends, platters and light lunches possibly pizzas and the like. We'll certainly be doing events, weddings and functions as well.
He said while the events and weddings are extravagant, the wine is the most important aspect for the team.
"Our focus is wine, the weddings and events are merely an add-on to a winery site wo our pure focus is wine," he said.
"We want to make sure the tasting experience is one that appeals to a really wide range of people.
"We do make a big, wide range of wines and because we've always offered them free for tasting without bookings, it just makes us really accessible."
Mr McLean said the Big Hill site would be open seven days a week, which he believed would be a point of difference between St Anne's and the other wineries in the area.
