The state government has requested Commonwealth assistance to help "double the distribution" of COVID-19 vaccines to GPs.
Acting premier James Merlino said authorities lodged a request with the federal government to help meet vaccine demands.
He said there was a significant increase in demand for both the Pfizer and AstraZeneca vaccines in the wake of the latest outbreak.
More coronavirus news:
"People are having to wait weeks, we have seen long queues, we have also seen gaps in access, particularly in regional Victoria and in our peri-urban areas," he said.
"This morning we have made a request to the Commonwealth, a request to double the distribution, double the number of AstraZeneca to our primary caregivers, our GP network.
"We agree with the AMA that our GPs can do double what they are doing. We want to see a doubling of the AstraZeneca vaccine to our GPs."
Mr Merlino said the government wanted to see more Pfizer allocations, starting with an additional 100,000 from mid-June, and some confidence in that supply.
A standalone quarantine centre for Victoria has taken a step forward with the federal government handing a memorandum of understanding to the state.
Mr Merlino said both parties had signed the memorandum of understanding.
"We are going to get cracking and deliver this facility," he said.
"What we know with hotel quarantine is it's done a very good job but it is not no risk. It is not a zero risk environment. Never has been and never will be.
"We have seen across our nation, 21 breaches in hotel quarantine. Our strong argument has been we must have a Howard Springs-like alternative, a purpose-built quarantine facility for our highest risk individuals."
Although a site is yet to be decided, Mr Merlino said proposed locations at Mickleham or Avalon would work.
The Commonwealth will pay for most of the building costs, while the state government will cover the ongoing costs of running the centre.
