The man had returned to his Woollert home on May 4, after quarantining in South Australia on return from overseas.
He developed coronavirus symptoms on May 8 and was tested on Monday.
Testing undertaken after a man returned to Melbourne confirmed the infection was acquired while in hotel quarantine in South Australia.

It comes as the state recorded zero new cases of community transmission on Wednesday.
One new case has been recorded in hotel hotel quarantine bringing the number of total active cases across the state to 19.
Authorities administered more than 7900 vaccinations on Wednesday with another 21,984 people tested for the virus.
