Paramedics transported three people from the scene of the crash on the Wimmera highway at Newbridge.
An Ambulance Victoria spokesperson said all three people were assessed at the scene with a woman in her 60s flown to The Royal Melbourne Hospital in a serious condition with injuries to upper and lower parts of her body.
A man in his 60s and a man in his 70s were both taken to Bendigo hospital in a stable condition with upper body injuries.

Police at the scene said one of the two people - a woman - initially trapped in their vehicle would be flown to hospital in Melbourne.
The second person being cut out of the car was a 72-year-old man.

A State Emergency Service spokesperson has confirmed SES crews are working to free two people from the same car following a head-on crash at Newbridge this morning.
"Both Marong and Bendigo unit volunteers are on scene," the spokesperson said.
The crash is on the Wimmera Highway near Bridgewater-Maldon Road and Twiggs Lane.
Victoria Police and Ambulance Victoria are on scene.
An Ambulance Victoria spokesperson said an air ambulance has been dispatched to the scene.
"Two people - a man and a woman - are being assessed at scene and an air ambulance has been dispatched," the said.

A person is trapped after a two-car crash on the Wimmera Highway near Newbridge.
Emergency services were called to the scene just before 8.30am.
A CFA spokesperson said one CFA unit and an SES crew were on scene with police and paramedics called.

"It appears there are two cars involved," the spokesperson said. "It also appears at this stage there is one patient that requires extraction (from one of the cars)."
