It comes after at least five people died on Victorian roads in the first two weeks of December.
A nearly three-week-long operation starting on Friday will target black spots and arterial roads with random alcohol and drug testing.
Bendigo Highway Patrol Acting Senior Sergeant Paul Gordon said the spate of death on the road was a concerning trend over the last few weeks.
Other news:
He said police expected more road users in the region, as people holidayed locally or interstate.
Acting Senior Sergeant Gordon urged people to take the appropriate rests on long trips, and plan these into their journeys.
He asked for patience on the roads, especially around larger vehicles such as caravans.
"Impatience is a big cause of road trauma on our roads," he said.
"We're asking people to be patient, take their time and share the roads equally with everyone else."
Acting Senior Sergeant Gordon said anyone planning to drink at an event should plan a lift home, or plan to take a taxi.
He said people could expect to see a high number of police on the road during the coming weeks.
