The 74-year-old man from Amphitheatre was driving north-east on Phelan Road when the crash occurred at 11.50am.
Police, CFA and Ambulance Victoria crews attended the scene.
"It appears there has been some sort of medical episode and the vehicle left the road and collided with a tree," Central Goldfields Highway Patrol acting sergeant Craig Walker said.
"He was the only person in the car. Paramedics arrived and commenced CPR but unfortunately the male was pronounced deceased at the scene."
