Catholic Education Sandhurst director Paul Desmond confirmed the cases today.
"I can confirm two more positive cases among staff at St Joseph's," he said.
"It's disappointing but not unsurprising. Experts would tell us it is a seriously contagious (virus) and when one case (is found) you likely end up with more."
The school closed last week after a staff member tested positive to coronavirus. It was scheduled to re-open on Wednesday but Mr Desmond said it was possible the closure could extend.
"(The school) has been thoroughly cleaned and students have been told it's (closed) at least until Wednesday," he said. "But (it will not re-open) until we are confident the clean is thorough and absolutely completed."

Mr Desmond said the school had already planned to go to remote learning.
"Principal Joan Coldwell has that in hand," he said. "We were going to remote learning anyway during cleaning period but it will extend."
In a letter to the school community, Ms Coldwell said she is waiting for advice from Bendigo Health about contact tracing.
"A decision has been made therefore for the school to remain closed for the following week," Ms Coldwell wrote.
"The school will not be able to provide supervision to children of essential workers. I understand this will inconvenience some families but I hope that you understand that the safety and wellbeing of all members of our community is a priority.
"From Monday, the students will be returning to remote learning."
Cleaning is also continuing at Catherine McAuley College campuses after a student tested positive to COVID-19 last week.
Holy rosary in Heathcote is also closed for cleaning after it closed due to a connection to one of the cases.
"Both (cleans) are going well," Mr Desmond said. "I have told company not to rush and be absolutely confident they've got it. There is no need to rush, it's important to get it right.
"In the meantime, we are certainly thinking of the three colleagues who are not well and (the student) at Catherine McAuley College. We're hoping and praying everyone is up and about soon."
