It is alleged the sports car was travelling at 140km/h in a 110km/h zone on the Calder Freeway about 1pm on Monday.
The 58-year-old driver also produced a positive preliminary oral fluid test.
The Cranbourne man is expected to be charged on summons with drug and speed related driving offences.
The intercept forms part of Operation Regal, a four-day road safety operation during the Queen's Birthday long weekend.
The state-wide operation aims to reduce road trauma, including the number of lives lost and injuries on our roads.
The operation involves all available personnel including local road policing units, general duties, other operational policing units, and centralised resources from Road Policing Command.
Operation Regal will finish on Monday at 11.59pm.
