Bendigo is one of 14 Victorian stores slated to reopen, with 35 stores returning nationally.
Myer announced it was closing all of its stores from March 30 to reduce the risk of spreading COVID-19.
Myer Bendigo store manager Peter Griffin is excited to welcome customers back to the store.
"We look forward to welcoming our loyal Bendigo customers back into store in a safe shopping environment," Mr Griffin said.
The Bendigo-born retailer has been reopening its stores progressively and on a staged basis, taking into account state government measures.
Myer said its change rooms will be open, with distancing and hygiene measures in place.
Sneeze guards will be rolled out at registers, while face masks and gloves will be made available to team members.
About 10,000 Myer staff across the country were stood down following its decision to close in late March.
Chief executive John King said the temporary trading halt was "one of the toughest decisions the company faced in its 120 years."
The iconic retailer's history spans more than a century in Bendigo.
In 1900, Myer brothers Sidney and Elcon opened the first Myer store in Bendigo.
There are now 60 stores operating in Australia.
This year, Myer's share price peaked at 52 cents on January 15 and plummeted to as low as 10 cents on March 23.
On Thursday, Myer shares closed at 27 cents on the ASX.
