Data released on Monday showed Campaspe Shire had seen three cases of the illness in total as of 11.59pm Sunday, but had no active cases.
It is not known whether the active case revealed on Tuesday is among these three cases, or instead a new case and the shire's total number has been revised.
The department will not comment on individual cases.
There are no active cases in Bendigo, which has not recorded a change in coronavirus figures since early April.
