"With restrictions being eased, now is the time to take bold action to restore confidence and jobs to our regional economies," opposition leader Michael O'Brien said.
Helping operators get back on their feet, upgrading tourism-related infrastructure and promoting Victorian destinations are among the proposed fund uses.
"Our country communities and small businesses have been hit by drought, bushfires, and now the coronavirus pandemic," Nationals leader and Member for Murray Plains, Peter Walsh said.
"Tens of thousands of regional Victorians who rely on their jobs in the tourism sector to pay the bills and put food on the table are counting on the Andrews Government to get them back to work and back in business."
The opposition suggested the state government scrap "self-promotional advertising" and redirect money into promoting regional Victorian tourism.
The state's spend on promoting its 'Big build' campaign last year was cited as an example of the government's "self-promotional advertising".
The opposition also called on the government to complete projects that would attract people to bushfire-affected areas of the state.
More to come.
