Police are investigating the circumstances around the fatal crash.
Investigators believe a small truck was travelling along Webb Road about 4am when the driver lost control.
Emergency services attended but the male driver died at the scene.
The Department of Transport has advised Webb Road will remain closed for some time.
Drivers should use Curr Road, the Midland Highway and Heathcote-Rochester Road as alternatives and allow extra time.
The road will re-open when investigations are complete and the scene is safe.
The death brings this year's road toll to 85. At the same time last year 112 people had died on the roads.
Anyone who witnessed the incident or who may have dashcam footage is urged to contact Crime Stoppers on 1800 333 000 or submit a confidential report at www.crimestoppersvic.com.au
